TECHNICAL SKILLS DNA: multiplex ASPE-based genotyping, DNA sequencing, multiplex ligation-dependent probe amplification, phage and plasmid library construction and screening, plasmid construction, polymerase chain reaction, oligonucleotide-directed in vitro mutagenesis, Southern blotting. RNA: TaqMan (quantitative RT-PCR), Northern blotting, primer extension, RNase protection, in vitro transcription. Protein: Western blotting, preparation of transcriptionally active extracts from HeLa cells, purification of over- expressed proteins from E. coli. Cell biology: tissue culture, transient transfections, creating stably transfected cell lines, antibody immunolocalization, in situ hybridization, light microscopy. DNA/protein interactions: electrophoretic mobility shift assays, DNAase I protection. In vitro target validation: small interfering (si)-RNA, antisense oligonucleotides, adenoviral vectors for gene delivery. Other: Utilization of genomic/molecular databases and software (ex. ENSEMBL, NCBI, UCSC genome browser, HGMD, Sequencher, Vector NTI, etc), yeast-2-hybrid screening, germline transformation of Drosophila melanogaster, Drosophila genetics, basic mouse handling including intraperitoneal injections and perfusions.
