A biocompatible biological component is provided comprising a membrane-mimetic surface film covering a substrate. Suitable substrates include hydrated substrates, e. g. hydrogels which may contain drugs for delivery to a patient through the membrane-mimetic film, or may be made up of cells, such as islet cells, for transplantation. The surface may present exposed bioactive molecules or moieties for binding to target molecules in vivo, for modulating host response when implanted into a patient (e. g. the surface may be antithrombogenic or antiinflammatory) and the surface may have pores of selected sizes to facilitate transport of substances therethrough. An optional hydrophilic cushion or spacer between the substrate and the membrane-mimetic surface allows transmembrane proteins to extend from the surface through the hydrophilic cushion, mimicking the structure of naturally-occurring cells. An alkylated layer directly beneath the membrane-mimetic surface facilates bonding of the surface to the remainder of the biological component. Alkyl chains may extend entirely through the hydrophilic cushion when present.

The present invention is directed to curable compositions that comprise a non-porous substrate, one or more cyanoacrylate polymerization initiators and a prepolymer composition comprising at least one liquid cyanoacrylate monomer or mixture of such monomers (solid or liquid) and/or cyanoacrylate oligomers. The non-porous substrate is a collection of individual particulates that are not bound, bonded or fixed to one another. At least one initiator is deposited on the surface of the individual particulates to form a plurality of initiator carriers. The prepolymer composition receives the plurality of initiator carriers to begin a controlled and consistent polymerization or curing of the liquid cyanoacrylate monomer in order to produce a biocompatible adhesive composition for use on living tissue. The present invention also provides for methods of making and using and devices for such a curable compositions, particularly in treating living tissue by applying to living tissue a biocompatible adhesive composition.

A method for producing a cyanoacrylate monomer, comprising the steps of (a) reacting cyanoacetate and formaldehyde or paraformaldehyde in the presence of a base or amine base catalyst to form an α-cyanoacrylate prepolymer or oligomer mixture; (b) subjecting the α-cyanoacrylate prepolymer or oligomer mixture to a substrate having free acid, acid derivative, or free aldehyde or ketone groups to remove the catalyst and to obtain a purified α-cyanoacrylate prepolymer or oligomer; and (c) subjecting the purified α-cyanoacrylate prepolymer or oligomer to depolymerization to obtain an α-cyanoacrylate monomer.

A medical adhesive composition comprising, based upon the total weight of the composition, from about 50 wt. % to about 99.9 wt. % of one or more α-cyanoacrylate monomers and from about 0.1 wt. % to about 5 wt. % of one or more non-steroidal anti-inflammatory drugs (NSAIDs). Suitable NSAIDs include ibuprofen and acetaminophen. The resulting compositions provide enhances fibroblast proliferation and reduced cytotoxicity compared to compositions that do not contain NSAID.
